چکیده

Fuel ethanol production from plant biomass hydrolysates by the baker’s yeast Saccharomyces cerevisiae is of great economic and environmental significance. Costeffective production of ethanol from plant biomass should not only be based on the readily fermentable starch and sucrose fractions of plant carbohydrates, but also on the much more resistant lignocellulosic fractions. The utilization of xylose, the second abundant monosaccharide in lignocelluloses, is essential for the efficient bioconversion of lignocellulosic fractions of biomass, yet still a technical barrier to the commercial fermentation and a major challenge in metabolic engineering.

Yeast metabolic engineering for hemicellulosic ethanol production.

Efficient fermentation of hemicellulosic sugars is critical for the bioconversion of lignocellulosics to ethanol. Efficient sugar uptake through the heterologous expression of yeast and fungal xylose/glucose transporters can improve fermentation if other metabolic steps are not rate limiting. Rectification of cofactor imbalances through heterologous expression of fungal xylose isomerase or modi...

Metabolic engineering for improved fermentation of pentoses by yeasts

The fermentation of xylose is essential for the bioconversion of lignocellulose to fuels and chemicals, but wild-type strains of Saccharomyces cerevisiae do not metabolize xylose, so researchers have engineered xylose metabolism in this yeast. Glucose transporters mediate xylose uptake, but no transporter specific for xylose has yet been identified. Over-expressing genes for aldose (xylose) red...

Improved ethanol production by a xylose-fermenting recombinant yeast strain constructed through a modified genome shuffling method

BACKGROUND Xylose is the second most abundant carbohydrate in the lignocellulosic biomass hydrolysate. The fermentation of xylose is essential for the bioconversion of lignocelluloses to fuels and chemicals. However the wild-type strains of Saccharomyces cerevisiae are unable to utilize xylose. Many efforts have been made to construct recombinant yeast strains to enhance xylose fermentation ove...

Metabolic engineering and classical selection of the methylotrophic thermotolerant yeast Hansenula polymorpha for improvement of high-temperature xylose alcoholic fermentation

BACKGROUND The methylotrophic yeast, Hansenula polymorpha is an industrially important microorganism, and belongs to the best studied yeast species with well-developed tools for molecular research. The complete genome sequence of the strain NCYC495 of H. polymorpha is publicly available. Some of the well-studied strains of H. polymorpha are known to ferment glucose, cellobiose and xylose to eth...

Characterization of the xylose-transporting properties of yeast hexose transporters and their influence on xylose utilization.

For an economically feasible production of ethanol from plant biomass by microbial cells, the fermentation of xylose is important. As xylose uptake might be a limiting step for xylose fermentation by recombinant xylose-utilizing Saccharomyces cerevisiae cells a study of xylose uptake was performed. After deletion of all of the 18 hexose-transporter genes, the ability of the cells to take up and...
